Do I need to put an weekly paid employee on hold if he didnt get paid for one week because he took unpaid leave? He will be back working but had the holiday booked before starting employment so took the week as unpaid.

Yes, when an employee isn't paid any money at all in a given pay run you need to mark them as on hold. This confirms to HMRC that the employee has no pay or liability in that period, but is still in employment.
